The Japan international joined Novak Djokovic, Rafael Nadal, Roger Federer, Alexander Zverev, Kevin Anderson, Marin Cilic and Dominic Thiem, one of the 8 players competing for November 11th.
Del Potro, reaching the top level of the season for the first three seasons this season, suffered a knee injury during the Masters championship last month.
"It's a shame that we are not able to compete in London," said Del Potro.
"It's a unique challenge and I've tried everything possible to improve the knee, the modification makes a good progress but we need more time."
Nishikori, 28, will make his fourth competitive appearance this term, reaching the semi-finals of 2014 and 2016.
